Working on the Protocol




This week in lab was about working out the specifics in the protocol for extracting DNA from egg white lysozyme.  The protocol given to me was a little vague and needed more details.  After repeating the experiment several times I was able to fill in the missing details.  It made things easier to follow and it helped me figure out the time frame for each DNA extraction.  Getting the egg white solution pH level to a 4 by adding acetic acid and 40% ethanol one drop at a time, was really time consuming.  Now, with the detailed protocol this process is a little faster and more efficient.  Tomorrow I will put the new protocols to test and hopefully see the improvements in time and consistency of the DNA extraction process.

  1. Hey Mary, I am SO happy to hear that you do not have to struggle with the annoying “one drop at a time” protocol (and I am pretty sure you are happy about it, too)! I know that took a great amount of time off of your hands and it honestly sucks because it did not seem to work! Fortunately, I am glad you were able to plug in the missing pieces and were able to make it work :)

  2. You and I both are happy about the protocol being updated so that I don't have to add one drop at a time of the Acetic Acid and 40% Ethanol. That procedure took forever! It was time consuming and not very efficient. Now I get to use a pipette and things can move along a lot faster. Such a simple detail makes a world of difference.
